Capsule Filling: Crosslinking in Gelatin Capsules

After being faced with a failed Phase I immediate-release oral solid dosage formulation of a pro-drug in gelatin capsules, scientists at Metrics were tasked with identifying the cause of the failure as well as with resolving the issue. After identifying the challenge of crosslinking, the process of two or more proteins chemically joining, the scientists concluded that the formulation is changed from gelatin capsules to hydroxypropyl methylcellulose (HPMC) capsules.
